Effect pf Immunization on the Phagocytosis of Mycobacterium by Macrophages of Rabbits


Abstract
Numerous studies on the interactions between tubercle bacilli and macrophages from normal or immunized animals were reported; however, the contributory significance of the rates or quantities of phagocytosis of tubercle bacilli by macrophages to resistance to tuberculosis were incompletely established. It will be worth-while to investigate further comparatively the phagocytic activity of normal or immunized macrophage against tubercle bacilli as one of criteria for degree of resistance to tuberculosis.
Thus, an experiment was planned, and performed to study the effect of immunization on the phagocytosis of mycobacterium, by macrophages of normal or immunized rabbits at different temperatures which might influence the metabolic activities of various cells.
Normal, or immunized rabbit macrophages by Mycobacterium tuberculosis(H3,Rv) or Mycobacterium bovis (BCG) were collected from peritoneal cavity, suspended in the CMF medium containing normal or immune sera, and mix with the suspensions of mycobacteria. The macrophage-mycobacteria suspensions were maintained for 90 minutes at 37¡ÆC and 33¡ÆC, and the appearance rates of macrophages with AFB and the percentages of the intacellular units to the total bacteria units were observed.
The results of the observation were summerized as follows;
1) The rates of phagocytosis of mycobacteria by macrophages were significantly increased by immunization with H37Rv or BCG strain, while showing the specificity immunologically by some degrees.
2) Higher percentages were showed in the intracellular units to the total bactera units of H37Rv strain than of BCG stirain in both normal and immune sera.
3) The most active phagocytosis was observed from 15 minutes to 60 minutes through incubation period.
4) No significant difference was observed under the designed conditions of experiments between 37¡ÆC and 33¡ÆC.
